摘    要:以紫外分光光度法作为庆大霉素含量测定手段,采用正交试验方法,对紫色小单孢菌产庆大霉素的发酵培养基及发酵条件进行优化,通过统计学分析,确定发酵培养基最优组合为:黄豆饼粉3.5%,CaCO30.5%,淀粉5.5%,CoCl20.0006%。优化的发酵条件为:种龄60 h,接种量12 mL,装液量50 mL/500 mL摇瓶。在此优化条件下添加一定量的氨基酸培养6 d,庆大霉素效价较优化前提高了85.5%。用紫外分光光度法与微生物法测定庆大霉素发酵液效价产生的误差小于4%,该方法可以用于教学实验中庆大霉素液体发酵培养基优化的快速筛选。

Abstract:Under the background of the operation experiment of gentamicin preparation and analysis,orthogonal test was used in micromonospora purpurea gentamycin fermentation to optimize the medium composition and cultivation conditions with UV spectrophotometry as a means of gentamicin potency determination.Through statistical analysis,the optimal fermentation conditions for gentamicin production were obtained as follows:soybean powder 3.5%,CaCO3 0.5%,starch 5.5%,and CoCl2 0.0006%.Research on the fermentation process showed that the best fermentation conditions were as follows:seed age was 60 h,inoculation volume was 12 mL,liquid volume in 500 mL and flask was 50 mL.Gentamicin potency of fermentation birth was determined after micromonospora purpurea inoculated 6 days with some aminophenol addition,and was 85.5% higher than initial value.The relative error of determination of gentamicin potency with UV spectrophotometry method used in this report is less than 4%.So,it could be applied in clipping screening of gentamicin fermentive medium optimization in experimental teaching.
