南方红豆杉枝叶总RNA的提取

摘    要:文章采用改良的异硫氰酸胍法提取南方红豆杉枝叶总RNA,通过甲醛变性凝胶电泳,可见3条比较明显的rRNA条带,其中28SRNA条带的亮度大约为18SRNA的2倍,说明总RNA样品的完整性良好。紫外吸收值测定,A260/A280比值介于1.7到2.0之间,A260/A230比值大于2.0,得到较为典型的RNA吸收值,说明酚类、蛋白质和无机盐等杂质已排除。此方法简便易行,能得到纯度高、质量好的RNA。

Abstract:General RNAs are extracted from branches and leaves of yews in the south by the improved method of guanidinium isothiocyanate, and three distinct rRNA bands can be observed by gelatin electrophoresis of dehyde denaturalization. The brightness of Band 28SRNA approximately doubles that of Band 18SRNA, indicating that General RNA specimen are integrited. By measuring ultraviolet absorption, the ratio of A260/A280 is from 1.7 to 2.0, and that of A260/A230 is over 2.0. These are typical RNA absorption values, indicating that all impurities have been removed, such as phenols, protein and inorganic salt, etc. RNAs of high purity and quality can be easily obtained by this simple method.
