原位RT-PCR检测胃癌中SNC66基因的表达

摘    要:在组织切片上进行原位RT-PCR扩增(in situ RT-PCR),并在扩增过程中掺入地高辛标记的尿嘧啶核苷酸(Dig-UTP),观察新基因SNC 66在胃癌组织与正常胃粘膜中的表达差异,以分析SNC 66与胃癌发生的相关性。结果发现,25个循环时,20例胃癌标本中有4例阴性不表达,16例呈强阳性表达;当循环数降至10时,则6例呈阴性,9例呈弱阳性表达,另有5例呈强阳性表达。表明SNC 66基因在胃癌组织中存在明显的表达降低或表达缺陷,SNC 66可能是一个消化道肿瘤的负相关基因。

Abstract:Paired tumor and normal paraffin - embeded tissues from 20 stomach cancer patients were tested by in situ RT -PCR in which the SNC 66 fragments were labeled with digoxingen to study the expressive phenominal of SNC 66 gene instomach cancer and investigate the relationship between the expression of SNC 66 gene and the genesis of the tumor. The re-suits showed that 4 were negative and 16 were strong positive in 20 cases when the PCR cycled 25 times,and 6 were negativeand 9 were weak positive while 5 were strong positive when the PCR cycled 10 tines. These results indicated that there was asignificant expression reduction of SNC 66 in stomach cancer tissues. SNC 66 should be further investigated as a digestivetube negative relative gene.
