蜘蛛染色体制片技术探索

摘    要:本文以多种蜘蛛不同的组织器官为实验材料,通过设置不同低渗处理时间和采用多种固定的方法,旨在找到蜘蛛染色体制片技术的最优化方案.结果表明:选取材料以蜘蛛处于发育中的生殖腺和幼蛛的附肢细胞为最好.在无水浴条件下,低渗时间对取血细胞为40分钟,对幼蛛取45分钟为最佳.用新鲜固定液进行固定,对第一次固定加以改进,采用高比例甲醇固定剂处理,有利于染色体伸展和染色体分散,得到理想的分裂相.

Abstract:In order to find the best smearing technology for spider chromosome,experiments have made by using different tissues and organs of diverse spiders as experimental materials,and designing different low-osmosis work-time and diverse fixed methods. Results indicate that the growing spider's reproduction gonad and young spider's appendage cell are thebest experimental materials; without water-bath, the low-osmosis work-time is 40 minutesfor blood cell (but 45 minutes for young spiders),and when high concentrated methanol isused as the first fixing-solution, it is of advantage for extension and dispersion of chromosome,and it is easy to gain the satisfactory divided-phase.
