应用RAPD技术对我国不同地区周期型马来丝虫DNA多态性的比较分析

摘    要:寄生虫分类学的研究对于寄生虫病原学,流行病学及防治等诸多方面具有学术及实际意义,已知在生物学上有差异的亚种或株,具有不同的致病力,我国不同地区马来丝虫的致病性、周期性、主要传播媒介媒价以及其他生物学特性等方面都有不同程度的差异,为了弄清这些差异的原因究竟是由于丝虫本身还是由于传播媒介或患者生活环境不同等因素所致,国内学者已应用电镜、酶组织化学及同工酶电泳分析等技术对我国不同地区的周期型马来丝虫进行了比较研究,为直接从基因结构上探讨各地区的丝虫是否有差异,本实验采用随机扩增多态性DNA(RAPD)分析技术,对我国湖北谷城(H)、四川乐山(S)及浙江安吉(Z)三个地区的周期型马来丝虫进行种下分型的研究探讨,本实验采用RAPD分析技术,对我国三个地区周期型马来丝虫进行DNA多态性比较分析,结果表明,不同地区的马来丝虫在基因结构上既有同源性(三个地区的马来丝虫有共享a、c带,H、S两地区的丝虫有a、c、d、e、f g 6条共享带),又存在差异(各地区丝虫的条带数不同),说明我国不同地区的马来丝早已存在种内分化。

Abstract:Parasitic taxology has practical application significance to the aetiology, epidemiology, prevention and control, treatment of paasitoses. Subspecies or strains on biolgy exist different pathogenicity. Pathogenicity, majoy vector of transmission and other biologic characteristic about periodic Brugia malayi (Bm) exist some differences. For make it clear that Periodic B.m from different endemic areas have differentiation or not, the scholars in our courtry have engaged in some studies in the field by electronmicroscope, enzymohistochemistry. isoenzyme electrophoresis analysis and other techniques. In order to explore it at gene level, We use random amplified Polymorphic DNA (RAPD) analysis to study subspecie taxonomy of Periodic BM from Gucheng in Hubei (H). Leshan in Sichuan (S), Anji in Zhe Jiang (Z).Using RAPD technique, we obtained the distinguishable bands patterns, which were partly the similar. It shows that Periodic B. m from different endemic areas have auto plidy while they exist differences too. Combining with past results by others methods, we can assume that Periodic Bm from different endemic areas have existed differentiation in intraspecies.
