摘    要:以籼稻的成熟胚为实验材料,经过成熟胚愈伤组织的诱导、继代培养,探索研究不同浓度激素和潮霉素对愈伤组织生长的影响以及农杆菌对转化效率的影响.结果显示,最适宜愈伤组织分化的6-BA浓度为2.0 mg·L-1,分化率达82.3%.潮霉素最佳浓度为30 mg·L-1时可以完全抑制愈伤组织的分化.该实验以杂交水稻的亲本籼稻不育系15S为材料,建立了农杆菌转化的体系,以期获得转PEPC基因的籼稻亲本,为杂种优势利用和生理育种的结合提供依据.

Abstract:With mature embryos of indica rice as experimental materials,the effect of different concentrations of hormone and hygromycin on callus and that of the agrobacterium on transformation efficiency were studied in this paper.The results show that the optimal differentiation medium is 6-BA 2 mg·L and and the regeneration frequency is 86.7 %.The optimal concentration with 30 mg·L of hygromycin can completely inhibit the differentiation of callus.In the experiment,the agro-bacterium tranformant system was established for obtaining the hybrid rice parent with PEPC gene,with a view to providing the basis for heterosis and physiological breeding.
