胶原诱导型大鼠类风湿关节炎模型的实验研究

目的：探讨大鼠类风湿性关节炎（RA）动物模型的制备、病理组织学观察及血清细胞因子IL-1、TNF-α检测。方法：以实验大鼠为对象,Ⅱ型胶原（CII）和完全弗氏佐剂（Complete Freund＇s Adjuvant,CFA）混合注射,不同时间点实验组大鼠（O、7、14、28、42d）所获得血样用于做IL-1、TNF-α检测,膝关节标本进行组织病理学研究。结果：模型组血清IL-1、TNF-α在急性期增高明显,与对照组差异有统计学意义（P〈0.05）。模型组关节组织标本观察到滑膜组织增生,其局部骨吸收破坏明显,软骨表层胶原纤维溶解、软骨细胞变性坏死,软骨表面凹凸不平。结论：采用异种CII与CFA制备的RA模型,可以用于RA组织病理学、发病机理及治疗方面的研究。

Experimental Research on Collagen-induced Rheumatoid Arthritis Rats Model

Objective：To establish rat rheumatoid arthritis model,observe pathlogic histology and detect products of IL-1,TNF-α in serum.Method ：The animal model was established by injecting CII and Freund＇s Complete Adjuvant into rats.The pathological changes of the rats ankle joint and IL-1、TNF-α in serum were studied respectively at the 0th,7th,14th,28th,42nd weeks after the animal models for joint had been established.Results：Assayed the rate of IL-1、TNF-α in serum at various time points,the values were increased obviously at acute phase,compared with normal group（P0.05）.Inflammation was presented in the ankle joint specimens,which included synovial hyperplasia,erosion into the articular cartilage and bone.The surface of the joint cartilages were rough,fibro of collagen broken and cartilage cell necrosis.Conclusion：By injecting the collagen protein II and Freund＇s Complete Adjuvant,we can get the animal models of rats rheumatoid arthritis（RA）,whose histological and clinical changes are similar to that of human,so it can be used to study mechanism of RA and treatment of RA.
