海马微注射L-精氨酸对大鼠空间学习记忆能力的影响

摘    要:目的:探讨大鼠海马CAl区微注射L-精氨酸(L—Arg)对大鼠空间学习记忆的影响。进而了解L—Arg的作用和机理,为深入研究该药物的神经生物学机制提供实验依据。方法:10只SD大鼠随机分为实验组(n=5,微量注射L—Arg0.5μl,浓度为50g/L)和对照组(n:5,微量注射生理盐水0.5μl),利用八臂迷宫对大鼠进行测试,记录不同药物注射下大鼠在八臂迷宫中的工作记忆错误和参考记忆错误。结果:大鼠海马区微注射生理盐水前后八臂迷宫实验3--作记忆错误分别为2±0.3次、2.4±0.2次,前后无显著变化(P〉0.05);注药前后参考记忆错误分别为6.2±0.3次、6.4±0.2次,前后无显著变化(p〉0.05)。实验测量结果对比发现,导管植入手术对大鼠空间学习记忆能力无明显影响;大鼠海马区微注射L-Arg前后测试时间分别为224.1s、223.8s,注射前后无明显差异(P〉0.05)。大鼠海马区注射L—Arg后工作记忆错误为1.9±0.3次,对照组为2.7±0.4次,实验组工作记忆错误次数明显降低(P〈0.05);注药后参考记忆错误分别为实验组5.3±0.4次、对照组6.5±0.5次,实验组参考记忆错误次数明显降低(P〈0.05)。结论:海马区微注射生理盐水对大鼠的学习记忆能力影响不大,海马区微注射L—Arg能提高大鼠的学习记忆能力。

Abstract:Abstract: Objective: To explore the influence about the capacity of space learning and memory through injection tiny L-arginine (L-Arg) in hippocampal CA1 area in rats, to get a better understanding about the function and mechanism with L-Arg, in the mean time, and to provide experimental basis for further research about the neurobiological mechanisms on the drug. Methods: using the eight-arms maze test to trained 10 SD rats with randomly divided into experimental group (n=5, microinjected L-Arg 0.5 μl/L, the concentration is 50g/L) and the control group (n=5, microinjected L-Arg 0.5 μl, the concentration is 50g/L), and record the times which the working memory errors and reference memory errors under injecting in different drugs in eight-arms maze . Results: with the injecting medicine in hippocampus the experimental group's working memory error is 1.9 ± 0.3 times, and the control group's is 2.7 ± 0.4 times; the experimental group's reference memory error is 5.3 ± 0.4 s, the control group's is 6.5± 0.5. The reference memory error with the experimental group and the control group under injecting the normal saline in eight-arms maze experiment is 2 ± 0.3 and 2.4 ± 0.2. The experimental group test time before and after microinjected L-Arg is 224.1s and 223.8s .The experimental group working memory error before microinjected medicine is reduce significantly than the control group (P〈0.05), and the control group is no obvious changes (P〉0.05). The control group working memory error before microinjected medicine is reduce significantly than the control group (P〈0.05), and the control group is no obvious changes (P〉0.05). The working memory errors and reference memory errors before and after microinjected the normal saline is no obvious changes (P〉0.05), and the test time is too no obvious changes (P〉0.05). Conclusion: the influence from microinjected the normal saline in hippocampal CA1 area is not obvious, and it can enhance the capacity which space learning and memory under microinjected L-Arg.
