摘    要:该实验通过SDS高盐法(常规法)、SDS高盐法(改良法)和Soil gDNA Kit法提取土壤微生物基因组DNA,用DNA Nanophotometer仪测定样本DNA的纯度和浓度,以确定最佳提取方法.结果表明,用SDS高盐法(常规法)、SDS高盐法(改良法)和Soil gDNA Kit法和土壤微生物基因组DNA的浓度分别为41.45 ng/μl、96.48 ng/μl和58.40 ng/μl,纯度(OD260/OD280)分别1.45、1.57和1.82.由结果可知,改良法提取DNA的浓度最高,而Soil gDNA Kit法的纯度最高,但其成本较高,因此,SDS高盐法(改良法)是节省成本且适合提取大批量土壤微生物基因组DNA的方法,今后在提取DNA过程中进一步优化,以提高其纯度.

Abstract:Soil microorganism DNA was extracted by SDS-high salt (general method)/SDS high salt method (im- proved method) and Soil gDNA Kit method in order to find out the best method in DNA extracting from a lot of samples. The results showed that the concentration of soil microorganism genome DNA with the three methods was 41.45 ng/μl, 96. 48 ng/μl and 58.40 ng/μl respectively, and the purification (0D260/ 0D280 )was 1.45, 1.57and 1.82 respectively. As suggested by the result, the concentration of genome DNA with SDS-high solt ( im- proved method) was the highest, while the purification of genome DNA with Soil gDNA Kit method was the best a- mong the three, but it cost more than the other two. It can be concluded that SDS-high salt (improved method) was a better method to extract DNA from a lot of soil samples which cost less. In the further research, the improved method will be optimized so as to improve the purification.
