穿山甲组织同工酶的研究

摘    要:采用聚丙烯酰凝胶电泳方法分析穿山甲(Manispentadactyla)的肝、骨骼肌、心、脑、腮腺、脾和肾7种组织的LDH,Pox,PGM和MDH同工酶.心脏组织LDH图谱以B亚基为优势,酶活性由LDH1到LDH5依次减弱;脑和鳃腺组织中的情况与此相类似,而骨骼肌中以A亚基为优势,酶活性由LDH5到LDH1依次减弱;肝和脾的情况与此相类似,肾组织中A亚基和B亚基的含量大致相等,酶活性以LDH3为最高.在所有的LDH酶谱上,LDH3都由两条含量相近的酶带组成.还发现穿山甲肝脏存在LDH5缺失的现象脑组织不表现Pox酶活性,而肌肉组织中有3条Pox酶带,心脏则只有2条酶带,其它组织只有1条酶带穿山甲各种组织的PGM图谱由PGM1和PGM2组成对于MDH而言,穿山甲的特点是m-MDH的活性远低于s-MDH.还讨论了穿山甲上述同工酶的特点和意义.

Abstract:This paper studied on isoenzymes (LDH, Pox, PGM, MDH) of seven kinds of tissues-heart, brain, kidney, spleen, liver, parotid gland and striated muscle by the way of vertical thin-layer polyacrylamide gel electrophoresis (PAGE), In heart, liver and spleen, B subunit of LDH was superiority, the activities of isoenzyme weakened successively from LDH1 to LDH5, in kidney, the centent of A subunit was the same as B subunit, the activity of LDH3 isoenzyme was the highest, The results showed that LDH isoenzyme bands lacked LDH5 in liver tissue of 2 pangolin. In addition, there were two LDH3 bands that had the same activity in all LDH isoenzyme patterns. Pox isoenzyme, in brain, had not activity,but, there were 3 isoenzyme bands in striated muscle, there were 2 isoenzyme bands in heart, there was a isoenzyme band in other tissues. The electrophorestic patterns of PGM isoenzyme showed that there were 2 isoenzyme bands: PGM-1 and PGM-2. MDH isoenzyme was divided into two parts: mitochondrial MDH (m-MDH) and solutility cytoplasm MDH (s-MDH), the activity of m-MDH in pangolin was greatly lower than that of s-MDH.This paper analysed the characteristics and meaning of isoenzymes in pangolin.
