拟南芥和大蒜中超氧化物歧化酶提取方法的比较

摘    要:用热变性法或磷酸缓冲液法从拟南芥植物中粗提取超氧化物歧化酶(SOD),再用丙酮法或硫酸铵分级盐析法纯化SOD.比较发现,热变性法粗提取的SOD回收率较高,而2种纯化方法的差异却无统计学意义;同时,还比较了用同一种方法所提取的拟南芥和大蒜之间的SOD活性,大蒜的SOD活性高于拟南芥.

Abstract:Thermal denaturation and phosphate buffer methods were applied to initial extraction of superoxide dismutase (SOD) from Arabidopsis thaliana plants, garnish with acetone method and ammonium sulfate classification salting out method were conducted to purify SOD. In result, the thermal denaturation method showed higher SOD recovery rate in extracting SOD comparing to the phosphate buffer method. SOD extracted using the two methods showed no difference in purification process. The conducted experiment concludes a SOD separation method of higher activity and higher purity.
