珙桐干种子总DNA提取方法的比较

摘    要:以珙桐干种子为材料,采用简易CTAB法、SDS法、高盐沉淀法与高盐低pH值法分别对珙桐干种子进行了总DNA提取效果的比较分析。结果表明,四种方法均能有效地提取较高质量的珙桐种子DNA,其中SDS法效果最好,获得的DNA纯度最高,产量最大,CTAB法次之,高盐沉淀与高盐低pH值法获得DNA质量相对较低,低于其它两种方法。获得最佳的DNA提取方法,可为PCR扩增,引物设计,筛选珙桐种子中的相关基因奠定基础。

Abstract:Four methods are used to extract total DNA from dry seeds of Davidia involucrata,including CTAB,SDS,high salt precipitation and high salt and low pH for studying their effects on DNA extraction.The result shows that high-quality DNA can be obtained.The effect of high-quality DNA extraction with the method of SDS is the best,and the CTAB method the second.But the quality DNA extraction with high salt precipitation method and high salt and low pH method is lower than other two methods.An optimized preparation method to obtain high-quality DNA is suitable for PCR,primer design,screening gene from seed of Davidia involucrata.
