鼻粘膜细胞变应性炎的病理通路及TWH的干预

摘    要:探讨TLR-NF-кB信号通路及NF-кB不同程度表达在变应性鼻炎(Allergic Rhinitis,AR)发病中的作用及雷公藤多甙(tripterygium wilfor-dii hook,TWH)的干预。应用卵清蛋白建造AR细胞模型,ELISA法测IgE含量,Real-Time PCR及Westernblot检测TLR及NF-кB的表达。结果,模型组IgE、TLR-4及NF-кB的表达较正常组均高;LPS组TLR-4及NF-кB的表达进一步增强,IgE的表达较模型组反而降低;DEX组及TWH组TLR-4及NF-кB、IgE的表达较模型组均降低(P<0.05);TLR4与NF-кB的表达高度相关。结果表明,TLR-NF-кB信号传导通路在AR发病中发挥作用,NF-кB的过高表达抑制IgE的生成,产生免疫偏移作用。

Abstract:The role of TLR-NF-κB signaling pathway and the differrent expression of NF-κB in pathogenesis of allergic rhinitis (AR)in rats and the intervine of TWH were investigated. OVA was provided to build the AR model. Realtime-PCR and Western-Blot were used to evaluate expression level of TLR4 and NF-κB, and ELISA was used to evaluate expression level of IgE. The expression of TLR4, NF-κB and IgE were significantly higher in M group than in Normal group (P〈0.05). The expression of TLR4 and NF-κB were significantly higher in LPS group than in others (P〈0.05), while the expression of IgE was lower than M group. The expressions of TLR, NF-I〈B and IgE were significantly decreased in group TWH and DEX. The expressions of TLR4 and NF-κB were highly correlated. The results showed that TLR-NF-κB Signaling Pathway played an important role in pathogenesis of AR. The higher expression of NF-κB inhibited the production of IgE, thus intervening the development of AR with immune deviation.
