顶空气相色谱法测定注射用头孢替唑钠中残留溶剂

目的:建立一种气相色谱法同时测定注射用头孢替唑钠中有机溶剂甲醇、乙醇残留量的方法.方法:采用标准加入法顶空自动进样,在非极性弹性毛细管柱上进行分离,效果良好.结论:方法重现性好,定量准确,便于操作.     

头孢匹胺钠致严重白细胞降低1例

目的通过对头孢匹胺钠致严重白细胞降低1例病案报道,为临床合理用药提供参考。方法病例回顾。30岁女性患者,胃镜检查为非萎缩性胃炎伴胆汁返流及十二指肠球部炎症,给予0．9％氯化钠注射液250mL＋注射用头孢匹胺钠2g,泮托拉唑80mg及维生素B60．1g,均为每日1次,静脉滴注治疗。结果用药2d后患者反馈头痛难忍,血常规WBC由8．95×10^9·L^-1降为2．5×10^9·L^-1,立即停用头孢匹胺钠,另两药继续3d,头痛缓解,5d后WBC恢复至正常水平。结论注射用头孢匹胺钠可致WBC严重降低,临床应密切注意,停止用药后可逐步恢复。

Expression of a bee venom phospholipase A2 from Apis cerana cerana in the baculovirus-insect cell

Bee venom phospholipase A2(BvPLA2) is a lipolytic enzyme that catalyzes the hydrolysis of the sn-2 acyl bond of glycerophospholipids to liberate free fatty acids and lysophospholipids.In this work,a new BvPLA2(AccPLA2) gene from the Chinese honeybee(Apis cerana cerana) venom glands was inserted into bacmid to construct a recombinant transfer vector.Tn-5B-4(Tn) cells were transfected with the recombinant bacmid DNA for expression.Sodium dodecylsulfate-polyacrylamide gel electrophoresis(SDS-PAGE) analysis revealed a double band with molecular weights of 16 and 18 kDa.Products of hexahistidine AccPLA2 fusion protein accumulated up to 5.32% of the total cellular proteins.The AccPLA2 fusion protein was cross reactive with the anti-AmPLA2(BvPLA2 of the European honeybee,Apis mellifera) polyclonal serum.The reaction resulted in a double glycosylation band,which agrees with the band generated by the native AmPLA2 in Western blot analysis.The PLA2 activity of the total extracted cellular protein in the hydrolyzing egg yolk is about 3.16 μmol/(min·mg).In summary,the recombinant AccPLA2 protein,a native BvPLA2-like structure with corresponding biological activities,can be glycosylated in Tn cells.These findings provided fundamental knowledge for potential genetic engineering to produce AccPLA2 in the pharmaceutical industry.

硫酸钡混悬液产品配方优化

利用正交试验探讨了六偏磷酸钠、枸橼酸钠、羧甲基纤维素钠（CMC）等助剂对医用硫酸钡混悬液产品稳定性的影响,得到的最优化质量浓度组合（g／ml）为六偏磷酸钠：CMC：枸橼酸钠-3：2：5。其中CMC因子对稳定性结果影响最大。该研究能为医用硫酸钡混悬液生产工艺的控制和改进提供有益的参考。     

Analysis of bulked segregants to identify molecular markers linked with cocoon weight and cocoon shell weight in the silkworm Bombyx mori L

Two silkworm strains viz, B20 A (high cocoon shell ratio) and C.Nichi (low cocoon shell ratio) were sib mated for 10 generations to determine the homozygosis. Both bulked segregant analysis(BSA) and near isogenic lines (NIL) studies were done to identify the RFLP markers closely linked to cocoon shell parameters. Three hundred and fifty-two random clones were identified as the low copy number sequence and used for identification of Restriction Fragment Length Polymorphic (RFLP) marker linked to cocoon weight and cocoon shell character. In the bulk segregant analysis, DNA from the parents (B20 A, C.Nichi), F1 and F2 progeny of high shell ratio (HSR) and low shell ratio (LSR) were screened for hybridization with the random clones. Polymorphic banding pattern achieved through southern hybridization with different probes indicated the probable correlation of polymorphism with high and low cocoon shell character which are possible landmarks in identifying the putative marker(s) for the cocoon shell character. Out of the 100 probes tried with parents, F1, F2 and their bulks, 10 probes were found to be closely linked to cocoon shell characters.     

巯基化合物键离解能的密度泛函和CBS-Q方法研究

利用密度泛函方法(B3LYP、B3PW91、B3P86、PBE0)结合6-311G**基组和完全基组(CBS-Q)方法计算了15种巯基化合物的键离解能。结果表明,CBS-Q和B3P86是计算巯基化合物键离解能的可靠方法。为了检验非局域BLYP方法是否适合研究巯基化合物的键离解能以及B3P86方法是否对基组敏感,利用BLYP/6-31+G*方法和B3P86方法结合6-31+G*、6-31+G**和6-311+G**基组,计算了7种巯基化合物的键离解能,并将所得结果与实验结果进行了比较,研究发现B3P86方法对基组不敏感。考虑到CBS-Q方法的计算成本和B3P86计算的可靠性,B3P86方法结合适中的或较大的基组可以准确计算巯基化合物中C-SH键的离解能。     

Science Letters:Assignment of CCR7 gene to chicken chromosome 27 by radiation hybrid panel mapping

The protein encoded by CC chemokine receptor 7 (CCR7) is a member of the G protein-coupled receptor family. This receptor was identified as a gene induced by the Epstein-Barr virus (EBV), and is thought to be a mediator of EBV effects on B lymphocytes. This receptor is expressed in various lymphoid tissues and activates B and T lymphocytes. It has been shown to control the migration of memory T cells to inflamed tissues, as well as stimulate dendritic cell maturation. To map the CCR7 gene in chicken chromosome, a 6 000 rads chicken-hamster radiation hybrid panel (ChickRH6) was used. PCR of samples from ChickRH6 revealed that the location of CCR7 gene is linked to the maker SEQ0347 (6 cR away) with LOD score of 16.6 and that the marker SEQ0347 is located on chromosome 27 at 27 cR of RH (radiation hydrid) map. We compared the corresponding human mRNA sequence with the predicted coding sequence of chicken CCR7 gene, and found that the assembled contig shared a high percentage of similarity with that of the human gene.     

In vitro remineralization of hybrid layers using biomimetic analogs

Resin-dentin bond degradation is a major cause of restoration failures. The major aim of the current study was to evaluate the impact of a remineralization medium on collagen matrices of hybrid layers of three different adhesive resins using nanotechnology methods. Coronal dentin surfaces were prepared from freshly extracted premolars and bonded to composite resin using three adhesive resins (FluoroBond II, Xeno-III-Bond, and iBond). From each tooth, two central slabs were selected for the study. The slabs used as controls were immersed in a simulated body fluid (SBF). The experimental slabs were immersed in a Portland cement-based remineralization medium that contained two biomimetic analogs (biomineralization medium (BRM)). Eight slabs per group were retrieved after 1, 2, 3, and 4 months, respectively and immersed in Rhodamine B for 24 h. Confocal laser scanning microscopy was used to evaluate the permeability of hybrid layers to Rhodamine B. Data were analyzed by analysis of variance (ANOVA) and Tukey’s honest significant difference (HSD) tests. After four months, all BRM specimens exhibited a significantly smaller fluorescent area than SBF specimens, indicating a remineralization of the hybrid layer (P≤0.05). A clinically applicable biomimetic remineralization delivery system could potentially slow down bond degradation.
